Harley Quinn appears Batman: Arkham City voiced by Tara Strong.She is shown wearing a biker-girl themed costume in this game, using a low-key version of her usual makeup, with heavy eyeshadow in lieu of her domino mask. This costume featured knee high wedge boots and a corset like top. She wears her hair in two pigtails on both sides of her head which she has dip dyed red and black. Harley had captured some doctors and police officers as hostages when Batman finds her in the clock tower

Selina Kyle is originally captured by Two-Faceintending to have her tried before a kangaroo court for pillaging his hideout. After being rescued by Batman, she attempts to locate stolen articles which were seized by the authorities upon her arrest. Catwoman, with the reluctant aid of Poison Ivy, eventually pilfers her loot from Hugo Strange's heavily-guarded vault, but sacrifices a chance to escape Arkham City in order to save Batman from the destruction of Joker's base. She returns at the end of the game, who looks at Batman carrying the Joker's dead body. The character's storyline, which culminates in a final showdown with Two-Face at an old natural history museum, has been estimated to be approximately ten percent of the Arkham City content.

Strange is eventually placed in charge of running Mayor Quincy Sharp's "Arkham City" project, a sprawling, open-air, prison district where incarcerated inmates are not kept in cells but allowed to wreak havoc on the streets

Mr Freeze
Mr. Freeze appears in Batman: Arkham City voiced by Maurice LaMarche as both an ally to Batman and a boss battle. After the Joker poisons Batman he has the Dark Knight find Mr. Freeze. Batman searches for him in the GCPD building (literally the coldest place in Arkham City), only to find his lab swarmed by the Penguin's men, who reveal that Penguin is holding Freeze hostage in the Cryus Pinkney National History Institute. It is revealed that the Penguin stole Freeze's gun and is holding GCPD officers hostage. Batman saves the hostages and Freeze from Penguin's men in the museum and asks Freeze for the cure to his ailment, only for Freeze to reveal he needs his suit, which Penguin possesses

The Penguin is redesigned without penguin-like mannerisms such as the waddle or the squawk-laugh, but retains his beaked nose and monocle (here depicted as the bottom of a glass, said to have been shoved into his face by a young man he was torturing after cheating at cards). He also wears what appears to be an electrolarynx strapped to or implanted in his neck. According to the Arkham City Stories included in the game, Cobblepot became trapped in the open-air prison perimeter of Arkham City after refusing to vacate his residence at the Iceberg Lounge (which was condemned as part of the new project).

Many years ago, Ra's and the League of Assassins started a project they called "Wonder City" which was to be a crime free and pro-environment utopia built underneath Gotham City. Ra's powers the city with the natural chemicals that were found underneath Gotham, concentrated in an area that would end up becoming the Lazarus Pit. The project fails, due to multiple reasons, though Ra's still did not give up on his dream of eliminating crime from Gotham. In present time, Batman requires his blood to create a cure for a disease that Batman, Joker, and multiple people of Gotham City are afflicted with, as the cure would degrade too quickly without the unique chemical formula of the Lazarus Pit and Ra's' prolonged exposure to it having turned his blood into an organic catalyst that would prevent degradation

He kidnaps Aaron Cash's Medical Protection Team and the medical personnel they were guarding, sets up death traps, and threatens to murder them all if Batman does not solve his riddles and save them. He also places down Riddler Trophies across Arkham City to be collected as part of the challenge. To access the location of Riddler's various trophies and riddles, Batman must interrogate his henchmen who are scattered throughout Arkham City and blending in with the gangs of Joker, Penguin, and Two-Face. At certain intervals, when Batman finds enough "challenges", the location of a room containing a hostage is revealed.
